擠出原理
Extrusion Principle
樣品經(jīng)過(guò)水化形成脂質(zhì)體后，一般會(huì)通過(guò)高壓均質(zhì)或者超聲的方式對(duì)樣品進(jìn)行初步均質(zhì)處理，處理后樣品粒徑和分布都會(huì)有所提高，但往往還不能達(dá)到所需要的理想要求。此時(shí)，將均質(zhì)后樣品由下圖中進(jìn)料口進(jìn)入，通過(guò)高壓氮?dú)鈱悠窂臑V膜中擠出，擠出后的樣品粒徑和分布都會(huì)進(jìn)一步提高。
After the sample is hydrated to form liposomes, the sample is usually homogenized by high-pressure homogenization or ultrasonic wave. After treatment, the particle size and distribution of the sample will be improved, but it can not meet the desired requirements. At this time, the homogenized sample enters through the feed port in the figure below, and the sample is extruded from the filter membrane by high-pressure nitrogen. The particle size and distribution of the extruded sample will be further improved.
支撐板
Support plate
支撐板和濾板主要起到支撐濾膜的作用。 由于磷脂有一定的相變溫度，所以在擠出之前需要對(duì)擠出器夾套通入恒溫水進(jìn)行預(yù)熱至一定溫度（如70℃），且在使用過(guò)程中需保持在此溫度。通常的脂質(zhì)體品種擠出之后粒徑能達(dá)到 100nm以下，PDI 能控制在 0.1 以下。
The supporting plate and filter plate mainly play the role of supporting the filter membrane.
